Hi. Can I replace my FC-T6010 48-36-26 Deore crank with this M6000 crank? Thanks.
Yes, you can as both are 10-speed Hollow Tec 2 systems.

Could this be used with a 9-speed setup?

The pedal thread on the drive side of my Shimano Deore 9 speed crank has stripped. Could I use this as an alternative, keeping everything else as is?
Generally NO this is 10 speed, you need to decide WHICH crank you have and try to replace it. As an absolute last resort you could buy VAR PE-04700 Pedal Thread Cutters for Helicoil Bushes (SJS Cycles part number : 35522) RT
