Will this work with Shimano Ultegra R8020, 11-speed? The hydraulic reservoir bleed screw has threaded and that’s what I’m looking to replace, is that part of the lever?

Sadly not- the levers are not replaceable on ST-R8020.

Do I need to purchase the entire lever or is the return shifter (the smaller inner plastic lever) part available separately?
